עלילת הסרט מתרחשת בסן פרנסיסקו של ימינו. ד"ר ויל רודמן (ג'יימס פרנקו), הוא מדען צעיר אשר מנסה לפתח תרופה לאלצהיימר באמצעות עריכת ניסויים גנטיים על קופים, בהם שימפנזה אותו הוא מכנה סיזר (אנדי סירקיס). התרופה יוצרת בהמשך זן חדש של קופים בעלי אינטליגנציה יוצאת דופן. הניסוי יוצא מכלל שליטה לאחר שסיזר משתחרר מהכלוב שלו, מפיץ את התרופה בקרב הקופים האחרים ומוביל אותם להתאגד ולמרוד בשלטון בני האנוש.
